Kevin McStay's side started well, with Cillian O'Connor slotting a penalty to give Mayo a three point lead at the break.
On a disappointing note in the first half, it was the other O'Connor - Diarmuid - who went off with injury early on.
Darragh Canavan had an immense second half, hitting the back of Robbie Hennelly's net to give Tyrone the lead.
The Ulstermen were able to hold on to the lead throughout, winning by four points in the end.
Next up for Mayo is Roscommon next weekend, where victory would be huge for either side in retaining their top flight status.
